The hotspot for all runners, inline skaters, bicylists and everybody else out to get some sunshine (or, at least some air) is the Lake Alster, north of downtown Hamburg. It is a loop of a little less than 10 km, basically following the shore of the lake and can be started at any point. If you travel by car, there is parking along Harvestehuder Weg, or take bus 109 to the Dammtor Station. There is one tricky part at the Eastern side, where the street Schöne Aussichten ends in a small park (a dead end). Instead, follow the street (now called Fahrhausstrasse), turn left at the next light, and you will be back on track after a few more meters. The loop takes you through parks and quiete neighbourhoods. For an extension, turn into Leipfad at the north end of the lake and run along the cannal there. By turning into Milchstrasse at the west shore and continuing on 3 blocks, you will be able to add some track workouts on the Hamburg University track located there (access is from Turmweg or Möllerstrasse).
